Έμπορος και γόνος της οικογένειας Ρούφου-Κανακάρη, η οποία κυριάρχησε στο εμπόριο της σταφίδας και της πολιτικής ζωής της πόλης, ήταν γιος του Αθανάσιου Κανακάρη (1760-1822) κτηματία, έμπορου,και προεστού της Πάτρας, ο οποίος πολέμησε κατα την επανάσταση του 1821 και ήταν αντιπρόεδρος του εκτελεστικού κατά την πρώτη περίοδο του αγώνα. Η διαφορά του ονόματος με τον πατέρα του οφείλεται στο ότι η οικογένεια Ρούφου, η οποία είχε Ιταλικές ρίζες από τη Σικελία και πριν έρθουν στην Ελλάδα ήταν ηγεμόνες της Καλαβρίας, στο πρωτότοκο αγόρι έδιναν το επώνυμο Κανακάρης και στα υπόλοιπα παιδιά το Ρούφος. Επί Καποδίστρια διατέλεσε διοικητής Ηλείας και Σύρου. Επίσης διετέλεσε σύμβουλος επικρατείας και υπουργός εξωτερικών. Εξελέγη δήμαρχος Πατρέων την περίοδο 1855-1858 και συμετείχε στην επανάσταση κατά του Όθωνα. Έγινε μέλος της αντιβασιλείας μαζί με τον Κωνσταντίνο Κανάρη και τον Δημήτριο Βούλγαρη απο τις 11 Οκτωβρίου 1860 εώς 19 Οκτωβρίου 1863, ενώ πρωθυπουργός το 1863 και το 1865-1866. Ήταν φιλομοναρχικός και εκπρόσωπος των κοτζαμπάσηδων της Αχαίας. Από την οικογένειά του διακρίθηκαν επίσης οι γιοι του Θάνος Κανακάρης, Γεώργιος Ρούφος που διατέλεσαν δήμαρχοι βουλευτές και υπουργοί, καθώς και οι γιοι του Θάνου Μπενιζέλος Θ. Ρούφος, ο οποίος διατέλεσε υπουργός εξωτερικών και ο Λουκάς Ρούφος που διατέλεσε βουλευτής. Πέθανε το 1868

Benizelos Rouphos (Greek Μπενιζέλος Ρούφος) was a Greek politician and Prime Minister.

Rouphos was born in Patra in 1795, a scion of the wealthy Rouphos-Kanakaris family. He was the son of Athanasios Kanakaris who fought during the Greek War of Independence.

During the rule of Ioannis Kapodistrias, Rouphos became governor of Elia. Later he would also serve as Foreign Minister.

In 1855, Rouphos was elected Mayor of Patra, a post he held for three years. When King Otto of Greece was exiled in 1862, Rouphos became one of three viceroys - along with Konstantinos Kanaris and Dimitrios Voulgaris - that held power from October 10, 1862 until October 19, 1863.

Rouphos served three times as Prime Minister of Greece. He died on March 18, 1868
