Coming from the academies of PAOK he scored his first Super League goal on June 20, 2020 against OFI in the play-offs in only his second appearance for the club.

In his first European game for PAOK on August 25, 2020 against Turkish side Besiktas Tzolis scored a brace, making him the third youngest player on the list (behind Stelios Pozoglou and Dimitris Salpingidis) to score in Europe. With 5 goals in Europe he is - aged 18 years old in December 2020 - already joined 10th in the list of top-scoring PAOK-players in European competitions.

Tzolis scored his first hattrick as a professional player on January 20, 2021 in a cup match for PAOK against Larisa.

On August 12, 2021 Tzolis was transferred for EUR 11,5 mio + bonusses + 17,5% resale to English Premier League side Norwich City. This was the biggest transfer fee PAOK ever got for a player. In his first season Tzolis made 14 appearances in the Premier League, most of them as a sub as Norwich was struggling against relegation and only three subs a game were allowed.

On July 22, 2022 Tzolis joined Dutch side FC Twente on loan to return to Norwich City on January 31, 2023.

As of March 27, 2023 he has been capped 13 times for the Ethniki, scoring one goal in his second game, a friendly against Cyprus.
